Short placebo-controlled acromegaly trial

Design

A double-blind randomized placebo-controlled trial assessed biochemical measures during 14 days of treatment.1

Population

The trial included 20 patients with acromegaly, with ten assigned to octreotide and ten to placebo.1

Outcomes reported

Mean growth-hormone and insulin-like growth factor I levels were significantly reduced during octreotide treatment, although response varied and two treated participants had no significant growth-hormone reduction.1

Limitations

The study was very small and lasted only 14 days, so it cannot establish long-term symptom control, clinical outcomes, or uncommon harms.1

MPOWERED maintenance trial

Design

A sponsor-funded, open-label randomized Phase 3 trial compared oral octreotide with injectable somatostatin receptor ligands after an oral run-in selected biochemical responders.2

Population

Ninety-two adults with acromegaly who had previously responded to injectable therapy and then completed the oral run-in as responders were randomized.2

Outcomes reported

Biochemical response was maintained by 91% of participants assigned to oral octreotide and 100% assigned to injectable therapy; the adjusted comparison met the prespecified noninferiority criterion.2

Limitations

The open-label trial randomized only oral-run-in responders, limiting generalizability to people who had not already tolerated and responded to both treatment forms.2
