REIMAGINE 1: combination in early diabetes
NCT06323174 ↗ · Combined treatment
What was tested?
Cagrilintide plus semaglutide versus placebo in adults whose type 2 diabetes was inadequately controlled with diet and exercise. ↗
Registered primary question: Change in HbA1c at week 40. ↗
What the reviewed report found
Journal year 2026 · Article summary reviewed 2026-09-15
Under the efficacy analysis, HbA1c fell by 1.8 and 1.5 percentage points in the combination groups versus 0.1 with placebo. Adverse events occurred in 79%, 75% and 66%, respectively, and were predominantly mild or moderate gastrointestinal events.↗
Keep in context: The primary outcome concerns blood glucose; weight change was secondary. These efficacy-estimand results should not be ranked directly against treatment-policy estimates from other populations. ↗↗

REIMAGINE 1: early type 2 diabetes
- Design in the report
- A blinded randomized placebo-controlled phase 3a trial assessed 40-week HbA1c change under NCT06323174.↗
- Participants in the report
- The trial randomized 189 adults whose type 2 diabetes was inadequately controlled with diet and exercise.↗
- Limitations
- The efficacy estimand differs from the treatment-policy analyses in the REDEFINE reports. These populations, endpoints and analyses should not be combined into a simple treatment ranking.↗
